The error message is not in a pop-up window
I have found from searching painstakingly through various forums (since your support is excruciatingly slow to respond), that for some reason the memory is getting unclicked in the project settings. When I click on it, the concordance search works, but the memory keeps getting unclicked. Why is this happening?
Unfortunately this "Object reference not set to an instance of an object" tells us nothing which is why Elisa asked for the stack trace. If you'd shown a screenshot of how the error surfaced if it's not a popup then we we could hopefully explain where to find the details that would be helpful.
Anyway... this isn't our support. It's a public forum. If you had technical support I'm sure it would be fast!

Hard to tell from this minimal information but a guess would be you have the TM stored in OneDrive with cloud only options and so Trados Studio cannot actually find it. Can you provide the full link to where the TM is actually held?
Or maybe you just don't have access to that memory at all?

It's not in the cloud it is on my company's server.

Potentially exactly the same problem. You are working with a file-based translation memory as if it's a server or cloud TM... which it is not. Is the other TM in the same location?

You are selecting an SDLTM that is not on your local machine. You are using a tool in the mode that is designed for a single user with resources on their own computer. If you want to use translation memories that are on a server then there are solutions for that to ensure that you don't have problems with files disappearing like this.
Just because you have put the file on your server doesn't mean it's always available to you when working with Trados Studio so any issues in connectivity can be the reason for a problem like this.
However, I cannot be conclusive about your problem as the only thing I have to go on is a screenshot showing one TM not connected (tells me nothing other than it's not connected at the moment) and the other one (possibly in the same location and possibly not) connected, a high level error message with no detail that I could use to see if the underlying cause is shown, and you telling me the TM is on a server somewhere.
The most obvious cause based on this is that Studio is unable to find that particular TM.
Don't forget that you are in a public forum here and are relying on the goodwill of others to help you. This is not a guaranteed support site. If you want someone to take over your computer and get all the information we're trying to extract from you then you need to take a support contract and the support team will surely be happy to assist.

I'm not sure I follow about the connectivity - my TM is file-based on our company's shared drive (G). Everything is working properly except the concordance search on source. I checked teh TM on that drive and it is being updated.

Confusing then... how is this possible when the TM is not enabled nor is the Update box?
Is the other TM you show also in the same location?
You'll also notice that there is no icon, another sign that the TM is not being seen at all. It was either there through virtue of a project template that did have access to it when the template was created, or it was there, the connection dropped, and it was lost.
I don't know how to give you any more detail on the error message. This is all I see:

ok - but it is still relevant to know why that one did work and the one you have a problem with did not. You're not helping here at all if you don't answer the questions :-)

My apologies if I haven't been clear so I will try to sum up the issues:
1. When I do a concordance search in Source, I get the error message I described earlier. When I do it in Target, it works fine.
2. My TM is saved on a shared drive at my work. It is not on the cloud or any other drive. I am not using or updating any other TM.
3. The TM is updating normally at the location on the shared drive.
So the only thing that is not working correctly is the concordance search in the source language.
