In a TM plugin when we call the SearchTranslationUnitsMasked function is there any way to know if we are in pre-translation or just doing interactive lookup in a document (or any other lookup mode)?

This information is not provided by the Translation Memory API but you could use Integration API to find when a document was opened. Using this event you might identify the moment a document was completely which should be after pre-translate hence the rest of the calls to the SearchTranslationUnitsMasked are interactive lookups
