We recently added a lot of user licenses for Content Manager. With only about 40 users to manage, the 'Settings-->Users' interface from web client wasn't too cumbersome. With over 100 users, this will be a lot more time consuming. Does anyone have any tricks on managing users from the server itself or does every manage users solely from the web client tool?

Can you elaborate more on the word "manage"? In general, the web client is UI oriented tool that allows one edit per user.
If you want to further automate then you need to script your query or changes. We provide a PowerShell automation module for this purpose. here are some links
