Have you got source and target language HTML files already?

If the answer to this question is yes then you can align them using the Studio alignment module which you'll find in the Translation Memories View.

If the answer is no then you need to translate them and as you translate them they will be added to your translation memory.

You said they are HTML files.  So no, you don't have to change the format.  Studio can read HTML files when aligning and also when translating.

I somehow feel as though I'm answering your questions, but it won't help because you don't know what questions to ask.  Can you perhaps explain exactly what you've got in terms of files and languages?

Sorry Paul. They are xml files.I have English and Italian xml files and i want to import them to align them and create a translation memory.

Is it correct??

ok.  XML files are fine, but since the translatable content in an XML could be set up in all kinds of weird and wonderful ways you might need to create a custom XML filetype for it.  It all depends how complex they are.  The best thing to do is this:

1. Open one of the source XML files for translation using the default settings in Studio.  There is a default XML that simply takes translatable content from every element in the file
2. Run a pseudotranslation using these options:

Once complete save the target file and inspect it.  All the translatable text will be replaced by dollar symbols.  This means anything missing will probably stand out like a sore thumb and is easily spotted.  If there is missing text, or text translated that you didn't want to be translated, then you will need to create a custom XML filetype for this before you do the alignment.
