Still struggling to get to grips with Trados after many years with DVX. I have a source segment with a word in bold at the beginning, I marked the target word as bold with ctrl + b but I can't make the remainder of the sentence unbolded.

You should better use CTRL+, called QuickPlace, NOT the bold option. When you use CTRL+B, the formatting might be wrong. If you use CTRL+, you get exactly the formatting which was in source.
Neither Studio nor any other CAT tool are formatting tools. Their concept is always to copy all the formatting from the source to target. Unbolding will work the same way - transfer the format from source to target via QuickPlace or - if you like mouse - by pressing CTRL and hitting the formatted text in source, while text in target is selected.
