I have been using Studio 2021 lately, but since this morning I cannot confirm any segments. I have tried uninstalling and re-installung, I have checked my user profile, I have renamed some folders hoping that Trados would recreate them on start-up. The problems remains. I can't find an API, can that be the problem?
Are you using Trados Studio desktop, or the online editor? Is this a cloud project?

It is Trados Studio desktop and it is an ordinary project, not a cloud project.
Is this the behaviour for all projects or just one?
Can you share a screenshot of the affected screen while this is happening?

This behaviour is for all projects. My temporary solution now is to manually copy the API from the Roaming folder to the Local folder. This API reappeared after re-installing Studio and opening a project. I cannot send you a screenshot (as Studio now works fine again).

With API i mean a little file called Sdl.ProjectApi.xml (User/AppData/Roaming/SDL/ProjectApi/Sdl.ProjectApi.xml). From my understanding this little file or a similar one should also be in User/AppData/Local/SDL/ProjectApi. I just copied the file from Roaming and pasted it randomly in Local (not even in the specific folder) and Studio works fine.
That's interesting... I don't have this file in my local folder at all. I have no idea how or even if this was the real reason for your problem going away. In Studio 2021 the folders in roaming/local were changed to cut down on the number of folders all over the place so comparing to an older version of Studio would not be productive.... if that's what you did?
Can you try removing that file and just see if the problem is still there? If it is we clearly need to investigate something here so your help would be appreciated.

For now I will focus on my translation projects and the approaching deadlines (low stress treshold, high stress level at the moment). I will try to reproduce the error next Monday or Tuesday and keep you up to date.

I have removed the api file, but cannot reproduce the original problem. I am 'confuzzled', I still do not know what caused Studio to behave like it did. Anyway, Studio works fine now so it is happy translating.
