The first thing to note is the interface is bilingual, not trilingual. So you can only ever do one language pair at a time in the view while translating.
Then, if your file is in Excel (you mentioned columns), then you can use the Multilingual Excel filetype to create a single project for all the languages you like. Then the application will populate the correct columns for you when you're done. So in this context the answer would be yes.
